Ravichandran Ashwin achieved the milestone of 500 Test wickets during Day 2 of the Rajkot Test, but unfortunately had to exit the match later that night.

Despite reaching the milestone, Ashwin had no choice but to pull out of the Test match. Late on the evening of February 16th, the BCCI made an announcement stating that he had to depart due to a family emergency. The BCCI also appealed for privacy for the all-rounder during this time.
On the same evening, BCCI Vice-President Rajeev Shukla expressed his well wishes for Ashwin’s mother, who had fallen ill. Shukla confirmed that Ashwin had to immediately leave for Chennai, making it impossible for him to continue participating in the Rajkot Test.
As a result of Ashwin’s withdrawal, India will now have to play with only 10 players. It is possible to bring in a substitute for the off-spinner, but this can only be done with the consent of Ben Stokes. This rule is outlined in Section 1.2.2 of the MCC Laws of Cricket. The potential replacements for Ashwin include Axar Patel and Washington Sundar.
